This is a working overview of enzymatic recycling, written for readers who want more than a one-paragraph summary but less than a textbook.
Reviewed 2026-01-02. Anything still debated is marked as such rather than presented as settled.
Common analytical approaches include enzymatic recycling assays, high-performance liquid chromatography, and mass spectrometry. Enzymatic recycling measures total glutathione after converting GSSG back to GSH, while separation methods can quantify GSH and GSSG separately. Derivatization may be used to improve detection or stability during analysis. LC-MS/MS offers high specificity and can distinguish glutathione from related thiols and adducts. Each method has different sensitivity, throughput, and susceptibility to interference, so method selection depends on the study question and sample matrix.
For solid glutathione reagents, storage at low temperature and protection from moisture and light are typical precautions. Aqueous solutions can oxidize over time, and pH affects stability; alkaline conditions generally promote thiol oxidation. Some protocols prepare fresh solutions, while others use antioxidants or chelators to limit metal-catalyzed oxidation. Purity and counterion content can vary among commercial preparations, affecting concentration calculations. Certificates of analysis and validated assays help verify identity and purity.
Commercial glutathione is produced by microbial fermentation or chemical synthesis, then purified. Reduced and oxidized grades are offered separately, with purity specifications often exceeding 98 percent. The compound appears in foods such as fresh fruits, vegetables, and meats, although cooking and processing can lower amounts. Oral, topical, and inhaled forms are discussed in research and consumer contexts, but absorption and tissue delivery remain active areas of study. Regulatory status varies by country and intended use.
Glutathione is a small sulfur-containing peptide built from glutamic acid, cysteine, and glycine. Its distinctive feature is a gamma-glutamyl bond between glutamate's side-chain carboxyl group and cysteine's amino group. This linkage resists ordinary peptidases and helps the molecule remain stable inside cells. The reduced thiol form, often abbreviated GSH, is the dominant intracellular species. The oxidized disulfide dimer, GSSG, forms when two reduced molecules link through their cysteine sulfur atoms. The balance between these forms is a common redox indicator.

Stability depends on pH, temperature, oxygen exposure, and trace metals. Aqueous solutions of reduced glutathione are susceptible to oxidation, especially when neutral or alkaline and exposed to air. Transition metal ions can catalyze thiol oxidation, so chelators and inert atmospheres are sometimes used in research settings. Standards are typically stored cold and desiccated, with limited freeze-thaw cycles. Questions remain about how closely in vitro stability data reflect the behavior of glutathione within intact cells and tissues.
Measuring glutathione requires attention to oxidation during sample handling, because GSH in biological samples can convert to GSSG or form mixed disulfides with proteins after collection. Acidic extraction, rapid cooling, and chelating agents are commonly used to limit such changes. Analytical methods usually distinguish free reduced glutathione, total glutathione, and protein-bound forms. Because these forms have different stability and reactivity, reported values depend heavily on the preparation protocol. No single preparation is universally suitable for every biological matrix or analytical goal.
For solid glutathione, storage conditions affect shelf life. The reduced form is typically kept cool, dry, and protected from air and light. Moisture can promote oxidation, while elevated temperatures accelerate degradation. Suppliers often specify storage at or below freezing, sometimes under inert gas. Solutions are less stable than powders and may require preparation shortly before use. Buffers and chelating agents can slow oxidation, but they do not eliminate it. Published stability data vary with matrix, pH, and container.
Quality control for glutathione focuses on identity, purity, and oxidation state. Certificates of analysis may report assay value, water content, and the presence of GSSG or other impurities. Chromatographic purity is often expressed as a percentage of peak area. Reference standards help laboratories compare results across instruments and batches. Because glutathione is a small, polar molecule, separation from cysteine, gamma-glutamylcysteine, and related thiols can be challenging. Verification often combines more than one analytical technique.
Measuring glutathione requires attention to sample preparation because the molecule oxidizes readily. Blood, tissue, and cell samples are often treated with acid to precipitate proteins and stabilize the thiol. Without such steps, GSH can convert to GSSG or form mixed disulfides during storage. Analytical methods include spectrophotometric assays, high-performance liquid chromatography, and mass spectrometry. Each approach has different sensitivity, specificity, and susceptibility to interference from related compounds in complex matrices.
Quantifying glutathione requires distinguishing GSH from GSSG and preventing oxidation during sample preparation. Common approaches include the enzymatic recycling assay, often called the Tietze method, which measures total glutathione after converting GSSG to GSH. HPLC with ultraviolet or fluorescence detection and LC-MS/MS can separate and quantify both forms, sometimes after derivatization of the thiol group. Blood, plasma, and tissue samples differ in matrix and baseline concentrations, so method validation must account for recovery, linearity, and interference. No single assay is universally standard.
Glutathione is most stable as a dry powder stored cool and dry, but its thiol group is readily oxidized in solution. Aqueous preparations at neutral or alkaline pH lose GSH faster because the thiolate form reacts with dissolved oxygen and metal ions. Acidic conditions, chelating agents, and oxygen exclusion can slow oxidation, while repeated freeze-thaw cycles promote degradation. Light exposure and trace metals also contribute to loss. Laboratories typically validate stability for their own matrices because degradation rates depend on pH, temperature, concentration, and container materials.
==== Liothyronine ==== Treatment with liothyronine (synthetic T3) alone has not received enough study to make a recommendation as to its use; due to its shorter half-life it would need to be taken more often than levothyroxine. Adding liothyronine to levothyroxine has been suggested as a measure to provide better symptom control, but this has not been confirmed by studies. In 2007, the British Thyroid Association stated that combined T4 and T3 therapy carried a higher rate of side effects and no benefit over T4 alone. Similarly, American guidelines discourage combination therapy due to a lack of evidence, although they acknowledge that some people feel better when receiving combination therapy. Guidelines by National Institute for Health and Care Excellence (NICE) discourage liothyronine. People with hypothyroidism who do not feel well despite optimal levothyroxine dosing may request adjunctive treatment with liothyronine. A 2012 guideline from the European Thyroid Association recommends that support should be offered concerning the chronic nature of the disease and that other causes of the symptoms should be excluded. The addition of liothyronine should be regarded as experimental, initially only for a trial period of 3 months, and in a set ratio to the current dose of levothyroxine. The guideline explicitly aims to enhance the safety of this approach and to counter its indiscriminate use. A 2014 guideline from the American Thyroid Association recommends against the use of liothyronine.
It induces excitotoxicity in rodents by overactivating NMDA and metabotropic glutamate receptors, leading to calcium overload and oxidative damage. In contrast, it targets glutamate-gated chloride channels in invertebrates, causing increased chloride permeability without affecting their excitatory glutamate receptors.
At the Game Developers Conference in March 2015, Valve and Taiwanese electronics company HTC unveiled SteamVR and the HTC Vive—a virtual reality platform and a virtual reality headset. The platform would be distinguished by its "Lighthouse" motion tracking system, where sensors on the headset and its included motion controllers read the position of two base station devices mounted in the play area. This would allow for "room-scale" VR experiences, where the player would not be required to remain in a stationary position in front of a camera and would be able to freely walk around the space. In November 2017, Microsoft added beta support for the SteamVR service for Windows Mixed Reality headsets. In June 2019, Valve released its own VR headset, known as the Valve Index, positioned as a higher-end device with wider field of view and higher refresh rate. They were accompanied by updated motion controllers, which are strapped against the user's palms and have sensors to detect input pressure and individual fingers. In November 2025, Valve announced the Steam Frame, a standalone VR headset.
=== Microdase === Microdase is a modified oxidase test used to differentiate Micrococcus from Staphylococcus by testing for the presence of cytochrome c. A positive result produces a dark color around the inoculant while negative result produces no color change.

Masterton and Slowinski used three criteria to describe the six elements commonly recognised as metalloids: metalloids have ionization energies around 200 kcal/mol (837 kJ/mol) and electronegativity values close to 2.0. They also said that metalloids are typically semiconductors, though antimony and arsenic (semimetals from a physics perspective) have electrical conductivities approaching those of metals. Selenium and polonium are suspected as not in this scheme, while astatine's status is uncertain. In this context, Vernon proposed that a metalloid is a chemical element that, in its standard state, has (a) the electronic band structure of a semiconductor or a semimetal; and (b) an intermediate first ionization potential "(say 750−1,000 kJ/mol)"; and (c) an intermediate electronegativity (1.9–2.2).
== Names == Calcitriol refers specifically to 1,25-dihydroxycholecalciferol. Because cholecalciferol already has one hydroxyl group, only two (1,25) are further specified in this nomenclature, but in fact there are three (1,3,25-triol), as indicated by the name calcitriol. The 1-hydroxy group is in the alpha position, and this may be specified in the name, for instance in the abbreviation 1α,25-(OH)2D3. It is also known as cholecalcitriol to emphasize its derivation from vitamin D3. Calcitriol is, strictly, the 1-hydroxylation product of calcifediol (25-OH vitamin D3), derived from cholecalciferol (vitamin D3), rather than the product of hydroxylations of ergocalciferol (vitamin D2). 1α,25-Dihydroxyergocalciferol (ercalcitriol) should be used for the vitamin D2 product. However, the terminology of 1,25-dihydroxyvitamin D, or 1,25(OH)2D, is often used to refer to both types of active forms of vitamin D. Indeed, both bind to the vitamin D receptor and produce biological effects. In clinical use, the differences are unlikely to have major importance. Calcitriol is marketed as a pharmaceutical for medical use under various brand names including Rocaltrol (Roche), Calcijex (Abbott), Decostriol (Mibe, Jesalis), Vectical (Galderma), and Rolsical (Sun Pharma).
As there are the E3 ligases containing HECT domains, in which they continue this 'transfer chain' by accepting once again the ubiquitin via another conserved cysteine and then targeting it and transferring it to the desired target. Yet in case of RING finger domain containing that use coordination bonds with Zinc ions to stabilize their structures, they act more to direct the reaction. By that, it's meant that once the RING finger E3 ligase binds with the E2 containing the ubiquitin, it simply acts as a targeting device which directs the E2 to directly ligate the target protein at the lysine site. Though in this case ubiquitin does represent other proteins related to it well, each protein obviously will have its own nuisances such as SUMO, which tends to be RING finger domain ligases, where the E3 simply acts as the targeting device to direct the ligation by the E2, and not actually performing the reaction itself such as the Ubiquitin E3-HECT ligases. Thus while the internal mechanisms differ such as how proteins participate in the transfer chain, the general chemical aspects such as using thioesters and specific ligases for targeting remain the same.

SNAP-7941 is a drug used in scientific research, which is a selective, non-peptide antagonist at the melanin concentrating hormone receptor MCH1. In initial animal studies it had promising anxiolytic, antidepressant and anorectic effects, but subsequent trial results were disappointing, and the main significance of SNAP-7941 is as the lead compound from which more potent and selective antagonists such as SNAP-94847 were developed, although it continues to be used for research into the function of the MCH1 receptor.
From April to May 2024, Rio Grande do Sul was hit by massive flooding, which also severely hit Porto Alegre. This was partly seen as, in addition to the sheer amount of precipitation, insufficient pumping of water, as trash and rubble had blocked many manholes in the city. Melo, during the flooding, affirmed that the city government knew that there was a possibility of strong rains and was not taken by surprise. He denied, however, that the city government had delayed its response and put the responsibility on the new round of floodings for the high volume, as well as the population, blaming them for the trash strewn throughout the city. During the floods, the Guaíba hit its highest ever historical level, leaving various parts of the city submerged and many people stranded. Due to the severity of the situation, the municipality set up 21 centers to receive thousands of people who were forced to leave their homes. The flooding was also marked by conflicts between Melo and the federal government, during which he criticized what he saw was the lack of support from them, despite having also delayed a formal emergency request for days. During this period, federal deputy Fernanda Melchionna and city councilman Roberto Robaina, both from PSOL, sued Melo. They both requested that the State Attorney's Office to start an investigation against Melo due to the fact that the municipal government had not sent the federal government the data of families in Porto Alegre hit by the floods.
== Genetic methods == The yeast two-hybrid and bacterial two-hybrid assays investigate interactions between artificial fusion proteins. They do not require isolation of proteins but rather use transformation to express proteins in yeast or bacteria, respectively. The cells are designed in a way that an interaction activates the transcription of a reporter gene or a reporter enzyme. These methods allow easy screening of interactions and libraries with high throughput (see two-hybrid screening).
== Industrial process design == In a typical scenario, an industrial process will use an extraction step in which solutes are transferred from the aqueous phase to the organic phase; this is often followed by a scrubbing stage in which unwanted solutes are removed from the organic phase, then a stripping stage in which the wanted solutes are removed from the organic phase. The organic phase may then be treated to make it ready for use again. After use, the organic phase may be subjected to a cleaning step to remove any degradation products; for instance, in PUREX plants, the used organic phase is washed with sodium carbonate solution to remove any dibutyl hydrogen phosphate or butyl dihydrogen phosphate that might be present.

Preanalytical factors such as sample type, time to processing, and stabilization method can change GSH and GSSG amounts. Analytical method and calibration also contribute to variation. Comparing absolute values across studies requires caution.
These assays typically measure total glutathione after oxidizing or reducing steps convert GSSG to GSH. A colorimetric or fluorometric signal is proportional to the recycling reaction. They generally do not report GSH and GSSG separately unless additional steps are used.
Solutions are often prepared fresh and kept cold, with protection from light and oxygen exposure. Chelating agents may reduce metal-catalyzed oxidation. Storage recommendations vary by buffer, pH, and concentration, so protocol-specific guidance should be followed.
It is a tripeptide rather than a full protein. Proteins generally contain many amino acids joined by alpha-peptide bonds, while glutathione has three residues and an unusual gamma-glutamyl linkage. That structure affects how enzymes recognize and break it down.
